FALCIGO 80mg Injection (Artemether)
Artemether is lipid soluble methyl ether of dihydroartemisinin. Artemisinin, also known as qinghao su, and its semi-synthetic derivatives are a group of drugs used against Plasmodium falciparum malaria. Artemisinin which has very rapid schizontocidal activity against blood forms of P. falciparum and P. vivax. After intramuscular administration, peak plasma concentrations are attained within about 6 hours.orArtemether has been reported to clear fever in severe falciparum malaria within 30-84 hours. It’s chemical formula is 3R.5as, 6R, 10S, 12R, 12aR)- Decahydro – 10-methoxy-3,6,9-trimethyl-3,12 epoxy-12H-pyrano[4,3-j]-1,2 benzodioxepin. Its molecular formula is C15H22O5and itsmolecular weight is 198.4.
Each 1 ml ampoule contains: Artemetether 80mg
Artemether is active against all plasmodia including those which may be resistant to other antimalarials. Artemether has very rapid schizotocidal activity. The schizotocidal activity of artemether is mainly due to the destruction of the asexual erythrocytic forms of P. falciparum and P. vivax. There is inhibition of protein synthesis during growth of trophozoites. There is no cross resistance of chloroquine. There is no rationaleat present for using artemether for chemoprophylaxis.
Pharmacokinetics data in human is sparse, with no data demonstrating the rate or extent of absorption or the systematic distribution of artesunate. After parental administration, artesunate is rapidly hydrolyzed to the active metabolite dihydroartemisinin. The oral formulation is probably hydrolyzed completely before entering the systematic circulation. Peak serum levels occur within one hour of oral dose of artesunate and persist for up to 4 hours. Following intravenous administration, elimination half –life of 45 minutes has been reported.
Dihydroartemisininhas a plasma elimination half-life of less than 2 hours. Which may show the development of resistance to artesunate.
Falcigo is an anti –malarial drug. It is indicated for the treatment of all kinds of malarias, including chloroquine resistance malaria and the first aid of fetal malaria.
Artemether is contraindicated in patient with hypersensitivity to artemether or other artemisinin compound. Artemether is not recommended in the first trimester of pregnancy because of limited data.
FALCIGO Injection SIDE EFFECT
Artemether has been remarkably well tolerated and appears less toxic than quinine or chloroquine, adverse effect inc include electrocardiogram abnormalities, gastrointestinal disturbance (nausea, abdominal pain, injection site pain, skin reactions and fever). Decreases in neutrophils and reticulocytes have been reported in some patients. There were no deaths or other side effect. No irreversible side effect were seen.
FALCIGO DOSAGE AND ADMINISTRATION
Injection: Intramuscular. Five days course with the dose of 480mg for adults: Once (80mg) daily for fie consecutive days with the first dose doubled. For children, the first dose is 3.2mg/Kg bodyweight, followed with 1.6mg/Kg bodyweight every time from the 2ndday to 5thday.
There is no experience with overdosage with artemether. There is no specific antidote known for the artemisinin derivaties.
Protect from light, heat and extreme cold. Do not refrigerate.
Keep out of the reach of children.
